Diawali dengan cerita masa kecil Logan "Wolverine" (Hugh Jackman) dan saudaranya Victor Creed “Sabretooth” (Liev Schreiber) . Pertemuannya dengan William Stryker (Danny Huston) pimpinan proyek Weapon-X akhirnya membuat kedua bersaudara ini saling bermusuhan.
Setelah kekasihnya dibunuh oleh Victor, Logan pun menuntut balas dan terjadilah pertarungan antara Logan dan Victor di mana pertarungan tersebut dimenangkan oleh Victor dengan menghancurkan cakar tulang yang dimiliki Logan.
